The p.G1527V variant (also known as c.4580G>T), located in coding exon 33 of the MYOM1 gene, results from a G to T substitution at nucleotide position 4580. The glycine at codon 1527 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces glycine with valine at codon 1527 of the MYOM1 protein (p.Gly1527Val). The glycine residue is moderately conserved and there is a moderate physicochemical difference between glycine and valine. In summary, this variant has uncertain impact on MYOM1 function. The available evidence is currently insufficient to determine its role in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
