ClinVar
Submissions by phenotype
not specified Uncertain:1
The p.Y326C variant (also known as c.977A>G), located in coding exon 5 of the MYOM1 gene, results from an A to G substitution at nucleotide position 977. The tyrosine at codon 326 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
MYOM1-related disorder Uncertain:1
The MYOM1 c.977A>G variant is predicted to result in the amino acid substitution p.Tyr326Cys. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0033% of alleles in individuals of South Asian descent in gnomAD (http://gnomad.broadinstitute.org/variant/18-3176085-T-C). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
Hypertrophic cardiomyopathy Uncertain:1
This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 326 of the MYOM1 protein (p.Tyr326Cys). This variant is present in population databases (rs773366188, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with MYOM1-related conditions. ClinVar contains an entry for this variant (Variation ID: 525006). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt MYOM1 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
