Variant summary: EIF2B5 c.584G>A (p.Arg195His) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 251394 control chromosomes. c.584G>A has been observed in multiple homozygous and compound heterozygous individuals affected with Leukoencephalopathy With Vanishing White Matter (e.g., Fogli_2002, Fogli_2004). It has also been observed to segregate with disease in related individuals. This variant is present in population databases (rs113994054, gnomAD 0.008%). This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 195 of the EIF2B5 protein (p.Arg195His). -
