The p.N1820S variant (also known as c.5459A>G), located in coding exon 39 of the DMD gene, results from an A to G substitution at nucleotide position 5459. The asparagine at codon 1820 is replaced by serine, an amino acid with highly similar properties. This alteration has been reported in an ostensibly healthy cohort (Vojinovic D et al. Eur J Hum Genet, 2015 Jun;23:837-43). Based on data from gnomAD, the G allele has an overall frequency of <0.01% (9/182449) total alleles studied, with 4 hemizygote(s) observed. The highest observed frequency was <0.01% (8/81342) of Eurpoean (non-Finnish) alleles. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
