The p.E74G variant (also known as c.221A>G), located in coding exon 4 of the RECQL4 gene, results from an A to G substitution at nucleotide position 221. The glutamic acid at codon 74 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
Baller-Gerold syndrome Uncertain:1
This sequence change replaces glutamic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 74 of the RECQL4 protein (p.Glu74Gly). This variant is present in population databases (rs759778382, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with RECQL4-related conditions. ClinVar contains an entry for this variant (Variation ID: 459389). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
