This missense variant replaces threonine with alanine at codon 506 of the CDH1 protein. To our knowledge, functional studies have not been reported for this variant. This variant has been observed in individuals affected with breast cancer, but was also observed in multiple healthy controls in a large breast cancer study (PMID: 32091409, 33471991). This variant has been identified in 8/282878 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The c.1516A>G (p.T506A) alteration is located in exon 10 (coding exon 10) of the CDH1 gene. This alteration results from a A to G substitution at nucleotide position 1516, causing the threonine (T) at amino acid position 506 to be replaced by an alanine (A). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
