Has not been previously published as pathogenic or benign to our knowledge; In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function; Reported in ClinVar as a variant of uncertain significance (ClinVar Variant ID# 581163; Landrum et al., 2016) -
not specified Uncertain:1
Variant summary: COL12A1 c.2569C>G (p.Gln857Glu) results in a conservative amino acid change in the encoded protein sequence. Four of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 7.2e-05 in 249506 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in COL12A1 causing Ullrich congenital muscular dystrophy 2 (7.2e-05 vs 0.0035), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.2569C>G in individuals affected with Ullrich congenital muscular dystrophy 2 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 581163). Based on the evidence outlined above, the variant was classified as uncertain significance. -
Inborn genetic diseases Uncertain:1
The c.2569C>G (p.Q857E) alteration is located in exon 13 (coding exon 12) of the COL12A1 gene. This alteration results from a C to G substitution at nucleotide position 2569, causing the glutamine (Q) at amino acid position 857 to be replaced by a glutamic acid (E). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
COL12A1-related disorder Uncertain:1
The COL12A1 c.2569C>G variant is predicted to result in the amino acid substitution p.Gln857Glu. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.014% of alleles in individuals of European (Non-Finnish) descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
