The p.S721C variant (also known as c.2162C>G), located in coding exon 17 of the BAP1 gene, results from a C to G substitution at nucleotide position 2162. The serine at codon 721 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
This missense variant replaces serine with cysteine at codon 721 of the BAP1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373).
