CFAP410 (HGNC:1260): (cilia and flagella associated protein 410) Four alternatively spliced transcript variants encoding four different isoforms have been found for this nuclear gene. All isoforms contain leucine-rich repeats. Three of these isoforms are mitochondrial proteins and one of them lacks the target peptide, so is not located in mitochondrion. This gene is down-regulated in Down syndrome (DS) brain, which may represent mitochondrial dysfunction in DS patients. [provided by RefSeq, Sep 2012]

This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 111 of the CFAP410 protein (p.Val111Met). This variant is present in population databases (rs555164150, gnomAD 0.006%). This missense change has been observed in individual(s) with retinitis pigmentosa and skeletal defects (PMID: 27548899, 33307614). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 428582). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on CFAP410 protein function. Experimental studies have shown that this missense change affects CFAP410 function (PMID: 27548899). For these reasons, this variant has been classified as Pathogenic. -
