The c.1289A>T (p.D430V) alteration is located in exon 9 (coding exon 9) of the RAD50 gene. This alteration results from a A to T substitution at nucleotide position 1289, causing the aspartic acid (D) at amino acid position 430 to be replaced by a valine (V). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
This sequence change replaces aspartic acid, which is acidic and polar, with valine, which is neutral and non-polar, at codon 430 of the RAD50 protein (p.Asp430Val). The frequency of this variant in the general population, 0.00083 (25/29982 chromosomes in South Asian subpopulation (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is higher than would generally be expected for pathogenic variants in this gene.
