This gene is found in a cluster of related genes on chromosome 14 and is thought to be involved in limb development. This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 125 of the SIX1 protein (p.Glu125Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of SIX1-related conditions (PMID: 21700001, 37479820). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 208361). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SIX1 protein function with a positive predictive value of 80%.
