RNA analysis indicates that this variant induces altered splicing and likely results in the loss of 4 and gain of 1 amino acid residue(s), but is expected to preserve the integrity of the reading-frame. This variant is present in population databases (rs748614767, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 540631). Studies have shown that this variant results in the activation of a cryptic splice site in exon 18 (Invitae). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The c.2019C>T variant (also known as p.G673G), located in coding exon 18 of the POLE gene, results from a C to T substitution at nucleotide position 2019. This nucleotide substitution does not change the at codon 673. However, this change occurs in the base pair of coding exon 18, which makes it likely to have some effect on normal mRNA splicing. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice donor site and will result in the creation or strengthening of a novel splice donor site. Based on the available evidence, the clinical significance of this variant remains unclear. -
