The p.I317T variant (also known as c.950T>C), located in coding exon 10 of the POLE gene, results from a T to C substitution at nucleotide position 950. The isoleucine at codon 317 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
