The p.D467E variant (also known as c.1401C>G), located in coding exon 14 of the NEBL gene, results from a C to G substitution at nucleotide position 1401. The aspartic acid at codon 467 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces aspartic acid, which is acidic and polar, with glutamic acid, which is acidic and polar, at codon 467 of the NEBL protein (p.Asp467Glu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with NEBL-related conditions. ClinVar contains an entry for this variant (Variation ID: 583115).
