This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 216 of the RIT1 protein (p.Asp216Asn). This variant is present in population databases (rs545031201, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with RIT1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1354441). Invitae Evidence Modeling incorporating data from in vitro experimental studies (internal data) indicates that this missense variant is not expected to disrupt RIT1 function with a negative predictive value of 95%.
