Variant summary: CHEK2 c.1259+8A>G alters a nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-05 in 249956 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for a pathogenic variant in CHEK2 causing Hereditary Breast And Ovarian Cancer Syndrome (4e-05 vs 0.00031), allowing no conclusion about variant significance. c.1259+8A>G has been reported in the literature in an individual affected with breast cancer without evidence for causality (example: Ackay_2020). The report does not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome.
