Variant summary: BRCA1 c.3055A>G (p.Ile1019Val) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251144 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3055A>G has been reported in the literature in a family affected with Hereditary Breast And Ovarian Cancer Syndrome without patient or cosegregation information (Judkins_2005). This report does not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. At least one publication reports experimental evidence evaluating an impact on protein function (Lu_2015). These results showed no damaging effect of this variant. The following publications have been ascertained in the context of this evaluation (PMID: 16267036, 15235020, 26689913). ClinVar contains an entry for this variant (Variation ID: 37504). Based on the evidence outlined above, the variant was classified as likely benign. -
