Variant summary
Our verdict is Uncertain significance. The variant received 4 ACMG points: 4P and 0B. PM2PP3_Moderate
The NM_014679.5(CEP57):c.704A>G(p.Gln235Arg) variant causes a missense change. The variant allele was found at a frequency of 0.000000688 in 1,453,888 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Synonymous variant affecting the same amino acid position (i.e. Q235Q) has been classified as Likely benign.

Mosaic variegated aneuploidy syndrome 2 Uncertain:1
In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with CEP57-related conditions. This sequence change replaces glutamine with arginine at codon 235 of the CEP57 protein (p.Gln235Arg). The glutamine residue is highly conserved and there is a small physicochemical difference between glutamine and arginine. -
