The protein encoded by this gene contains a DNA/RNA helicase domain at its C-terminal end which suggests that it may be involved in both DNA and RNA processing. The p.N1004D variant (also known as c.3010A>G), located in coding exon 8 of the SETX gene, results from an A to G substitution at nucleotide position 3010. The asparagine at codon 1004 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the supporting evidence, this variant is unlikely to be causative of autosomal dominant juvenile amyotrophic lateral sclerosis; however, its contribution to the development of autosomal recessive spinocerebellar ataxia with axonal neuropathy is uncertain. -
