Variant summary
Our verdict is Uncertain significance. The variant received 0 ACMG points: 1P and 1B. PP3BP4
The NM_016038.4(SBDS):c.671T>C(p.Ile224Thr) variant causes a missense change. The variant allele was found at a frequency of 0.0000285 in 1,613,906 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. I224M) has been classified as Uncertain significance.

The p.I224T variant (also known as c.671T>C), located in coding exon 5 of the SBDS gene, results from a T to C substitution at nucleotide position 671. The isoleucine at codon 224 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
