Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with hypertrophic cardiomyopathy (PMID: 25611685). This variant has been identified in 15/264942 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The P69L variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The P69L variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The P69L variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. The variant allele was found at a frequency of 6e-05 in 233582 control chromosomes in the gnomAD database, including 1 homozygotes. The observed variant frequency is approximately 2.4 fold of the estimated maximal expected allele frequency for a pathogenic variant in PRKAG2 causing Cardiomyopathy phenotype (2.5e-05). To our knowledge, no occurrence of c.206C>T in individuals affected with Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 45708). Based on the evidence outlined above, the variant was classified as likely benign. -
