In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This sequence change replaces alanine with threonine at codon 85 of the EFEMP2 protein (p.Ala85Thr). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and threonine. -

The p.A85T variant (also known as c.253G>A), located in coding exon 3 of the EFEMP2 gene, results from a G to A substitution at nucleotide position 253. The alanine at codon 85 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
