This variant disrupts the p.Asp1989 amino acid residue in NOTCH1. Other variant(s) that disrupt this residue have been observed in individuals with NOTCH1-related conditions (PMID: 25132448), which suggests that this may be a clinically significant amino acid residue. This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 1989 of the NOTCH1 protein (p.Asp1989His). -
