This variant is present in population databases (rs121908828, gnomAD 0.006%). This missense change has been observed in individuals with clinical features of TMEM127-related condition (PMID: 21156949, 28384794). ClinVar contains an entry for this variant (Variation ID: 126971). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
