Primary ciliary dyskinesia Uncertain:2
The c.1127C>G (p.T376S) alteration is located in exon 7 (coding exon 7) of the CCDC40 gene. This alteration results from a C to G substitution at nucleotide position 1127, causing the threonine (T) at amino acid position 376 to be replaced by a serine (S). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
This sequence change replaces threonine, which is neutral and polar, with serine, which is neutral and polar, at codon 376 of the CCDC40 protein (p.Thr376Ser). This variant is present in population databases (rs372392201, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with CCDC40-related conditions. ClinVar contains an entry for this variant (Variation ID: 407775). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
