This sequence change replaces histidine, which is basic and polar, with asparagine, which is neutral and polar, at codon 177 of the GNMT protein (p.His177Asn). This variant is present in population databases (rs121907889, gnomAD 0.07%), and has an allele count higher than expected for a pathogenic variant. This missense change has been observed in individual(s) with glycine N-methyltransferase deficiency (PMID: 11810299). It has also been observed to segregate with disease in related individuals.
