This variant is present in population databases (rs28937906, gnomAD 0.0009%). This missense change has been observed in individuals with autosomal recessive Charcot-Marie-Tooth disease (PMID: 12499475, 14561495, 18812441). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 4195). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
