The variant allele was found at a frequency of 0.00019 in 236140 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in PLEKHG5 causing Distal Spinal Muscular Atrophy, Autosomal Recessive 4 (0.00019 vs 0.0011), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.2525G>A in individuals affected with Distal Spinal Muscular Atrophy, Autosomal Recessive 4 and no experimental evidence demonstrating its impact on protein function have been reported. The p.R842Q variant (also known as c.2525G>A), located in coding exon 19 of the PLEKHG5 gene, results from a G to A substitution at nucleotide position 2525. The arginine at codon 842 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
