The c.4382G>A (p.R1461Q) alteration is located in exon 28 (coding exon 27) of the ALS2 gene. This alteration results from a G to A substitution at nucleotide position 4382, causing the arginine (R) at amino acid position 1461 to be replaced by a glutamine (Q). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Infantile-onset ascending hereditary spastic paralysis    Uncertain:1 
This sequence change replaces arginine with glutamine at codon 1461 of the ALS2 protein (p.Arg1461Gln). The arginine residue is moderately conserved and there is a small physicochemical difference between arginine and glutamine. This variant is present in population databases (rs758889641, ExAC 0.003%). This variant has not been reported in the literature in individuals with ALS2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
