The p.L58F variant (also known as c.172C>T), located in coding exon 1 of the FANCM gene, results from a C to T substitution at nucleotide position 172. The leucine at codon 58 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces leucine, which is neutral and non-polar, with phenylalanine, which is neutral and non-polar, at codon 58 of the FANCM protein (p.Leu58Phe). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with FANCM-related conditions.
