This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 93 of the NEUROG3 protein (p.Arg93Leu). This variant is present in population databases (rs121917838, gnomAD 0.003%). This missense change has been observed in individuals with congenital malabsorptive diarrhea (PMID: 16855267, 26541772). ClinVar contains an entry for this variant (Variation ID: 5323). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects NEUROG3 function (PMID: 31178402, 31805014).
