A literature search was performed for the gene, cDNA change, and amino acid change (where applicable). No publications were found based on this search. This variant is present in population databases (rs759124595, gnomAD 0.09%). This missense change has been observed in individual(s) with high hyperdiploid acute lymphoblastic leukemia (PMID: 31102422). ClinVar contains an entry for this variant (Variation ID: 408156). Variant summary: FANCE c.397C>T (p.Leu133Phe) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00016 in 251220 control chromosomes, predominantly at a frequency of 0.001 within the Latino subpopulation in the gnomAD database. The observed variant frequency within Latino control individuals in the gnomAD database is approximately 2 fold of the estimated maximal expected allele frequency for a pathogenic variant in FANCE causing Fanconi Anemia phenotype (0.00048). To our knowledge, no occurrence of c.397C>T in individuals affected with Fanconi Anemia and no experimental evidence demonstrating its impact on protein function have been reported.
