The c.583+3A>G vari ant in TMEM43 has not been previously reported in individuals with cardiomyopath y and was absent from large population studies. This variant is located in the 5 ' splice region. Computational tools do not suggest an impact to splicing and at >15 mammals carry a guanine nucleotide (G) at this position. Moreover, guanine is the second most common nucleotide at this position in the consensus splice se quence, supporting that this change may be tolerated. In summary, while the clin ical significance of the c.583+3A>G variant is uncertain, these data suggest tha t it is more likely to be benign. -
