The PALB2 c.1684+6del intronic change results in a deletion at the +6 position of intron 4 of the PALB2 gene. Algorithms that predict the impact of sequence changes on splicing indicate that this variant likely does not affect splicing, but to our knowledge these predictions have not been confirmed by RNA studies. To our knowledge, this variant has not been reported as pathogenic in individuals with PALB2-associated conditions. In summary, the evidence currently available is insufficient to determine the clinical significance of this variant. It has therefore been classified as of uncertain significance. -

This sequence change falls in intron 4 of the PALB2 gene. It does not directly change the encoded amino acid sequence of the PALB2 protein. It affects a nucleotide within the consensus splice site.
