The p.V723I variant (also known as c.2167G>A), located in coding exon 14 of the BRIP1 gene, results from a G to A substitution at nucleotide position 2167. The valine at codon 723 is replaced by isoleucine, an amino acid with highly similar properties. This alteration was observed in 1 of 64523 individuals with a personal history of breast cancer (Easton DF et al. J Med Genet, 2016 05;53:298-309). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -

Variant summary: BRIP1 c.2167G>A (p.Val723Ile) results in a conservative amino acid change located in the ATP-dependent helicase, C-terminal domain of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.8e-06 in 261718 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2167G>A has been reported in the literature in one individual affected with Breast Cancer (Easton_2016). This report does not provide unequivocal conclusions about association of the variant with Breast Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Four clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -

ClinVar contains an entry for this variant (Variation ID: 230464). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt BRIP1 protein function with a negative predictive value of 80%.
