The p.H1227R variant (also known as c.3680A>G), located in coding exon 19 of the BRIP1 gene, results from an A to G substitution at nucleotide position 3680. The histidine at codon 1227 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This variant is denoted BRIP1 c.3680A>G at the cDNA level, p.His1227Arg (H1227R) at the protein level, and results in the change of a Histidine to an Arginine (CAT>CGT). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. BRIP1 His1227Arg was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Histidine and Arginine share similar properties, this is considered a conservative amino acid substitution. BRIP1 His1227Arg occurs at a position that is not conserved and is not located in a known functional domain (Cantor 2011). In silico analyses predict that this variant is unlikely to alter protein structure or function. Based on currently available evidence, it is unclear whether BRIP1 His1227Arg is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. -
