A similar protein in mouse is a component of a protein complex termed biogenesis of lysosome-related organelles complex 1 (BLOC-1), and binds to alpha- and beta-dystrobrevins, which are components of the dystrophin-associated protein complex (DPC). Mutations in this gene are associated with Hermansky-Pudlak syndrome type 7. This gene may also be associated with schizophrenia. This sequence change replaces threonine, which is neutral and polar, with alanine, which is neutral and non-polar, at codon 264 of the DTNBP1 protein (p.Thr264Ala). This variant is present in population databases (rs753618520, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with DTNBP1-related conditions.
