This variant was seen in one patient with ARVC. Testing was performed at Invitae. Given the weak gene-disease association and lack of case data we consider this variant a variant of uncertain significance and we do not feel it is suitable for assessing risk in healthy relatives ("predictive genetic testing"). The MYPN gene is associated with autosomal recessive nemaline myopathy and there's some preliminary evidence associating it with autosomal dominant DCM, HCM and RCM (Purevjav et al. 2012). The variant has not been reported in the literature (per PubMed and Google). Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change. The variant is not listed in the Genome Aggregation Consortium Dataset (gnomAD; http://gnomad.broadinstitute.org/), which currently includes variant calls on >140,000 unrelated individuals of African, Asian, European, Latino, and Ashkenazi descent. The average coverage at that site in gnomAD is 90x in exomes and 33x in genomes. -

Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). This sequence change replaces asparagine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 668 of the MYPN protein (p.Asn668Ile). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MYPN-related conditions. ClinVar contains an entry for this variant (Variation ID: 477745). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
