This variant is absent in 120888 control chromosomes. In addition, one clinical diagnostic laboratory classified this variant as uncertain significance. The variant of interest has not, to our knowledge, been reported in affected individuals via publications; nor evaluated for functional impact by in vivo/vitro studies. Because of the absence of clinical information and the lack of functional studies, the variant is classified as a variant of uncertain significance (VUS) until additional information becomes available. -

The p.G114R variant (also known as c.340G>A), located in coding exon 2 of the RAD51C gene, results from a G to A substitution at nucleotide position 340. The glycine at codon 114 is replaced by arginine, an amino acid with dissimilar properties. In multiple assays testing RAD51C function, this alteration showed an abnormal read-out (Hu C et al. Cancer Res, 2023 Aug;83:2557-2571). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -

The RAD51C c.340G>A variant is predicted to result in the amino acid substitution p.Gly114Arg. To our knowledge, this variant has not been reported in the literature in affected individuals. However, in a functional study of variants of uncertain significance in RAD51C from public databases, p.Gly114Arg showed impaired activity in homology-directed repair (HDR) assays, conferred sensitivity to cisplatin and olaparib, and disrupted formation of protein complexes of RAD51C (Supplementary Tables 1-3 in Hu et al. 2023. PubMed ID: 37253112). This variant has been reported in 0.0080% of alleles in individuals of African descent in gnomAD and has been interpreted as variant of uncertain significance in ClinVar (https://www.ncbi.nlm.nih.gov/clinvar/variation/187382/). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -

ClinVar contains an entry for this variant (Variation ID: 187382). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt RAD51C protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects RAD51C function (PMID: 37253112). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
