The p.D159E variant (also known as c.477T>G), located in coding exon 3 of the RAD51C gene, results from a T to G substitution at nucleotide position 477. The aspartic acid at codon 159 is replaced by glutamic acid, an amino acid with highly similar properties. In multiple assays testing RAD51C function, this alteration showed a normal read-out. (Hu C et al. Cancer Res, 2023 Aug;83:2557-2571). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
This missense variant replaces aspartic acid with glutamic acid at codon 159 of the RAD51C protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with ovarian cancer or breast cancer (PMID: 34326862). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
