This protein is also localized to the endoplasmic reticulum lumen, however, its capacity for calcium-binding may be absent or much lower than other family members. This gene is specifically expressed in the testis, and may be required for sperm fertility. Mutation in this gene has been associated with familial hypertrophic cardiomyopathy. The c.1114G>C (p.E372Q) alteration is located in exon 9 (coding exon 9) of the CALR3 gene. This alteration results from a G to C substitution at nucleotide position 1114, causing the glutamic acid (E) at amino acid position 372 to be replaced by a glutamine (Q). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
