This missense change has been observed in individual(s) with DPM3-related congenital disorder of glycosylation (PMID: 28803818, 31266720). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is present in population databases (rs778481307, gnomAD 0.004%). This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 14 of the DPM3 protein (p.Leu14Pro). -
