DICER1 (HGNC:17098): (dicer 1, ribonuclease III) This gene encodes a protein possessing an RNA helicase motif containing a DEXH box in its amino terminus and an RNA motif in the carboxy terminus. The encoded protein functions as a ribonuclease and is required by the RNA interference and small temporal RNA (stRNA) pathways to produce the active small RNA component that represses gene expression. This protein also acts as a strong antiviral agent with activity against RNA viruses, including the Zika and SARS-CoV-2 viruses. Alternative splicing results in multiple transcript variants. DNA sequence analysis of the DICER1 gene demonstrated a sequence change, c.3496G>T, in exon 21 that results in an amino acid change, p.Val1166Phe. This sequence change does not appear to have been previously described in individuals with DICER1-related disorders and has also not been described in population databases such as ExAC and gnomAD. The p.Val1166Phe change affects a poorly conserved amino acid residue located in a domain of the DICER1 protein that is not known to be functional. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Val1166Phe substitution. Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Val1166Phe change remains unknown at this time. -

This variant has not been reported in the literature in individuals affected with DICER1-related conditions. This sequence change replaces valine with phenylalanine at codon 1166 of the DICER1 protein (p.Val1166Phe). The valine residue is weakly conserved and there is a small physicochemical difference between valine and phenylalanine. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.V1166F variant (also known as c.3496G>T), located in coding exon 20 of the DICER1 gene, results from a G to T substitution at nucleotide position 3496. The valine at codon 1166 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
