The c.619C>G (p.L207V) alteration is located in exon 5 (coding exon 5) of the DNAAF1 gene. This alteration results from a C to G substitution at nucleotide position 619, causing the leucine (L) at amino acid position 207 to be replaced by a valine (V). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
This sequence change replaces leucine with valine at codon 207 of the DNAAF1 protein (p.Leu207Val). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and valine. This variant has not been reported in the literature in individuals affected with DNAAF1-related conditions. ClinVar contains an entry for this variant (Variation ID: 242165). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
