This sequence change replaces arginine with serine at codon 345 of the CCDC39 protein (p.Arg345Ser). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and serine. This variant is not present in population databases (ExAC no frequency). This missense change has been observed in individual(s) with Tetralogy of Fallot, combined with cleft lip and palate (PMID: 28230599). ClinVar contains an entry for this variant (Variation ID: 216832). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
