The variant (rs587777426) is rare (<0.1%) in a large population dataset (gnomAD v4.1.0 7/1614172 total alleles, 0.0004%, 0 homozygotes) and has been reported in ClinVar (Variation ID: 135636). Two bioinformatic tools queried predict that this substitution would be damaging to the protein. The leucine residue at this position is evolutionary conserved across most of the species assessed. We consider the clinical significance of c.9616C>A in SYNE1 to be uncertain at this time.
