A literature search was performed for the gene, cDNA change, and amino acid change (where applicable). Publications were found based on this search. The evidence from the literature, in combination with allele frequency data from public databases where available, was sufficient to determine this variant is unlikely to cause disease. Therefore, this variant is classified as likely benign.
This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 211 of the LRRK2 protein (p.Ala211Val). This variant is present in population databases (rs112794616, gnomAD 0.04%), and has an allele count higher than expected for a pathogenic variant. This missense change has been observed in individual(s) with Parkinson disease (PMID: 17222106, 18213618, 19405094, 21885347, 29248340, 30598256). ClinVar contains an entry for this variant (Variation ID: 39223). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
