Variant summary: USH2A c.5666A>G (p.Asp1889Gly) results in a non-conservative amino acid change located in the Fibronectin type III domain (IPR003961) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 250586 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.5666A>G has been reported in the literature in individuals affected with Usher related conditions with a non-informative genotype (example: Aparisi_2014, Sanchez-Navarro_2017, and PereaRomero_2021). These report(s) do not provide unequivocal conclusions about association of the variant with Usher Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
