The protein is found in the basement membrane, and may be important in development and homeostasis of the inner ear and retina. Mutations within this gene have been associated with Usher syndrome type IIa and retinitis pigmentosa. Multiple transcript variants encoding different isoforms have been found for this gene. The USH2A c.7494T>A; p.Ser2498Arg variant (rs760977747), to our knowledge, is not reported in the medical literature or gene-specific databases. The variant is reported in the ClinVar database (Variation ID: 228223) and is listed in the general population with an overall allele frequency of 0.003% (8/250,694 alleles) in the Genome Aggregation Database. The serine at this position is weakly conserved, with several other mammalian species with arginine at this position, and computational algorithms (PolyPhen-2, SIFT) predict this variant is tolerated. However, given the lack of clinical and functional data, the significance of the p.Ser2498Arg variant is uncertain at this time. -
