TSHB (HGNC:12372): (thyroid stimulating hormone subunit beta) The four human glycoprotein hormones chorionic gonadotropin (CG), luteinizing hormone (LH), follicle stimulating hormone (FSH), and thyroid stimulating hormone (TSH) are dimers consisting of alpha and beta subunits that are associated noncovalently. The alpha subunits of these hormones are identical, however, their beta chains are unique and confer biological specificity. Thyroid stimulating hormone functions in the control of thyroid structure and metabolism. The protein encoded by this gene is the beta subunit of thyroid stimulating hormone. Mutations in this gene are associated with congenital central and secondary hypothyroidism and Hashimoto's thyroiditis. Alternative splicing of this gene results in multiple transcript variants. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251286 control chromosomes. c.145G>A has been reported in the literature as homozygous in two individuals affected with Isolated thyroid-stimulating hormone deficiency and has been shown to segregate with disease in one family (Hayashizaki_1989). These data indicate that the variant is likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in loss of mutant beta protein to form TSH polypeptides with alpha peptide when microinjected into Zenopus oocytes (Hayashizaki_1989). The following publication have been ascertained in the context of this evaluation (PMID: 2792087). ClinVar contains an entry for this variant (Variation ID: 12684).
